A solid-phase extraction column separates compounds through interactions with a packed sorbent. A method may retain the target for later elution, or retain interferences while collecting the target in the flow-through. Unlike particle filtration, separation depends on adsorption and desorption. Sorbent chemistry, capacity and solvent composition therefore belong to the analytical method rather than a universal operating recipe.
| Selection factor | What to compare |
|---|---|
| Retention mechanism | Reversed-phase, normal-phase and ion-exchange media use different interactions with molecular polarity and charge. |
| Bed capacity | Account for competing matrix components as well as the target load. |
| Elution strength | Solvent strength or charge changes release retained compounds but may affect downstream analysis. |

Control flow and inspect relevant fractions for target breakthrough.
Measure recovery, blanks and matrix effects before concentration or solvent exchange.
Its main mechanism is chemical retention; frits may incidentally trap particles.
Breakthrough, excessive washing, incomplete elution and later concentration losses can produce similar results.
